Histone H3 and H1 levels are reduced relative to genomic DNA content. Histone H3 and genomic DNA content were measured simultaneously in uninfected T-cell lines (CEM, Jurkat and Molt4) and HTLV-1 infected T-cell lines (SLB-1, HuT102 and C81) using flow cytometry. H3 levels were measured using a primary antibody directed against the C-terminal domain and an Alexa-488 labeled secondary antibody. DNA content was measured using propidium iodide. (A) Mean histone H3 immunofluorescent intensity was traced from G1 through G2/M by defining a narrow window on the DNA histogram and plotted against the genomic DNA content for the corresponding cell population defined by the window.
