SUMO interacting motifs are important for HIV-1 restriction by rhesus macaque TRIM5α. CRFK, TE671 and HeLa cells were transduced with a retroviral vector encoding wild type rhTRIM5α, SIM1mut, SIM2mut and SIM3mut or an empty vector (EV) control. A. Western blot assay showing the expression of the FLAG-tagged rhTRIM5α, using an anti-FLAG antibody, GAPDH was used as a loading control. B-C. Cells stably expressing the various rhTRIM5α proteins or an EV control were infected with an HIV-1 firefly luciferase reporter virus pseudotyped with VSV-G envelope. 48 hours after infection luciferase activity was measured. Representative experiment of four independent experiments for CRFK (B), HeLa (C) and TE671 (D) cells. E.
