LEDGIN treatment during virus production hampers infectivity and increases the quiescent fraction of residual provirus. Various cell lines were infected with OGH virus produced in the presence of LEDGIN CX014442 (concentrations indicated on x-axes) and analyzed by flow cytometry 3 days post infection. Upper panels: Dose–response curves showing the percentage of total infected mKO2+ cells (quadrant B + C, Fig. 2b) with increasing concentration of LEDGIN CX014442. Data represent averages of duplicates with standard deviation from a representative experiment in each cell line. In total five experiments were performed in Jurkat cells, ten in SupT1 cells and three in MT-4 cells. Three different virus dilutions are depicted in various shades of red. Middle panels: Dose–response curve showing the percentage of productively infected (eGFP+, mKO2+; quadrant B in Fig. 2b) cells with increasing concentration of CX014442.
