(B, C and E) Mapping of the interaction between APH-2 and Tax2B. 293T cells were transiently transfected with the indicated expression plasmids. Cellular extracts were prepared and immunoprecipitated with the indicated antibodies (IP) 48 hours post-transfection. The presence of the proteins of interest in the immunoprecipitates was visualized by Western blotting using the indicated antibodies (WB). (D) The non-canonical bZIP domain of APH-2 is required to inhibit Tax2B-mediated AP-1 transcription. 293T cells were transfected with the pAP-1 luciferase construct and the indicated expression vectors. Luciferase and Renilla activities were measured 48 hours after transfection. The average of at least three different experiments is shown. The expression levels of the transfected proteins were analysed by Western blot using the indicated antibodies.
