5A Knockdown of PAF1, CTR9 and RTF1 results in enhanced HIV-1 RT products - early and late (0-16 hr pi) and proviral DNA ( Alu - gag , 48 hr pi). Control wells containing inhibitors were negative for HIV-1 DNA. HIV-1 DNA copies are normalised to genomic GAPDH and presented per million cells. Results are mean ± SD of a representative experiment performed in duplicate. 5B Transfection of a PAF1-HA expression construct into HeLa-CD4 cells results in increased levels of PAF1. The PAF1-HA clone possesses an HA and an orange tag (~30 kDa, accounting for the 110 kDa band detected). 5C Over-expression of PAF1-HA resulted in a decrease in FFU/ml following challenge with HIV-189.6wt. Results are representative of multiple experiments and are mean ± SD. 5D Over-expression of PAF1-HA results in decreased RT products - early and late (8 hr pi) and proviral DNA (integrated, 48 hr pi). HIV-1 DNA copies are normalised to genomic GAPDH and presented per million cells. Results are mean ± SD of a representative experiment.
