In vitro RAL susceptibility of wt and mutated HIV-2 INs. Mutations were introduced in the HIV-2 N1 background by mutagenesis. (A) Comparison of strand transfer activity in the presence of RAL of wt (circle), G140S (square) and G140S/Q148R (triangle) mutants. (B) Comparison of strand transfer activity in the presence of RAL of wt (circle), N155H (triangle), Y143C (square) and N155H/Y143C (inverted triangle) HIV-2 INs. Strand transfer reaction was carried using a 32P-labeled oligonucleotide mimicking the preprocessed substrate and 200 nM IN, in the presence of increasing concentrations of RAL at 37°C. Activity is expressed as a % of control without drug. Experiments were performed two times.
