Phenotypic identification of thymic DC. (A) DC were gated based on CD123, CD11c^lo or CD11c^hi expression. (B) A major plasmacytoid (pDC; HLA-DR^intCD123^hiCD11c^-; 77.1%); a smaller myeloid (HLA-DR^int/hiCD123^-CD11c^lo; 20.4%); and a minor myeloid-related M-DC8⁺ (HLA-DR^int/hiCD123^-CD11c^hiM-DC8⁺; 2.5%) DC population were identified. Expression of myeloid antigens (CD1c and CD14), adhesion and co-stimulatory molecules (CD11c and CD11b), HIV-1 receptors (CD4, CXCR4 and CCR5), maturation markers (CD83 and CD86), C-type lectin receptors (DC-SIGN, DEC-205 and MR), CD123, HLA-DR and M-DC8 was determined.
