The mutant tax alleles were subcloned into a lentivirus vector, HR'CMV-SV-puro. The activity of each mutant to transactivate HTLV-1 LTR and NF-κB was determined by cotransfection of an HTLV-1 LTR luciferase construct or an E-selectin luciferase construct with each HR'CMV-tax mutant construct into 293T cells. The HTLV LTR (solid bars) and NF-κB (open bars) reporter activities of each mutant were normalized against those of the wild-type tax and expressed as % wild-type activity.Back to article page
