Nef increases levels of HIV-1 produced from SupT1 cells by a CD4 independent mechanism.A) SupT1 cells (1 × 107cells) were electroporated with 10 μg of plasmids directing the expression of wild type HIV-1NL4-3 and mutant HIV-1NL4-3ΔNef proviruses. 2 days later, supernatants and cells were collected and p24 levels were measured by p24 capture ELISA (top panel). Viruses from cell supernatants were concentrated by ultracentrifugation. Viruses and cell lysates were processed for western blotting (WB) with α p24 antibodies (bottom panel).
