The highly conserved KR box within the Np24 protein of M-PMV is required for viral replication. Culture media from COS-1 cells transfected with nothing (Black), wild-type M-PMV proviral DNA, pSARM4 (Red), or ΔKKPKR proviral DNA (Blue) were collected and assayed for RT activity. HOS cells were infected with equal amounts of virus (normalized by RT activity). Virus spread in HOS cells was measured by RT assays at 2, 4, 6, 8, 10, 12, and 14 days post infection.
