HIV-1 TAR miR-TAR-5p and miR-TAR-3p target different subsets of host genes. A) Minimal free energy (MFE) was calculated for a subset of mRNAs selected from the list produced by human miRTAR algorithm. B) HEK 293 cells were co-transfected with U6-shNEG (control) or U6-TAR and a vector containing the Renilla luciferase (Rluc) reporter gene usptream of three copies (3X) of the natural TAR-miRNA binding sites from the selected mRNAs (n = 1 to 3 experiments, in duplicate).
