Time of drug addition does not modify the effects of PIs and RTIs on HIV-1 cell-to-cell spread: HIV-1 infected Jurkat cells (donors) were mixed target cells in the presence of A) of LPV (14 μM) and C) DRV (12 μM) without prior pre-incubation of donor cells with drugs (time of addition t = 0 h). DNA was extracted at various times post-mixing and qPCR was performed as described in Figure 1. Data show mean of triplicates and error bars represent the standard deviation of the mean (SD). For RTIs, uninfected Jurkat cells (targets) were pre-incubated with B) TDF (20 μM) or D) AZT (10 μM) for 24 h prior to mixing with infected donor cells. DNA was extracted at various times post-mixing and qPCR was performed.
