Specific preference for CREB by Tax. (A) An example of CAT assay. HeLa cells were transfected with pU3RCAT alone (lane 1), pU3RCAT plus Tax expression plasmid pIEX (lane 2) or pU3RCAT plus pIEX plus increasing amounts (5 to 10 μg) of plasmids expressing the indicated dominant-negative proteins (lanes 3–10). D-Threo-[dichloroacetyl-1-14C]-chloramphenicol was as used as substrate in the CAT assay. (B) Influence of dominant-negative proteins on Tax activation. The cells received pU3RCAT (red) or pKB-SV40-CAT (blue) only (group 1), pU3RCAT/pKB-SV40-CAT plus Tax-expressing plasmid pIEX (group 2) or pU3RCAT/pKB-SV40-CAT plus pIEX plus plasmids expressing the indicated dominant-negative proteins.
