Mutations at the CA-SP1 junction confer DSB sensitivity to SIV, as revealed by single-cycle infection assays. Wild type and mutant SIV viruses were harvested from transfected 293T cells cultured in the presence of the indicated concentrations of DSB. Panel A: immunoblot analysis of viral lysates using a monoclonal antibody specific for SIV CA. Panel B: effects of DSB on SIV infectivity. Viral supernatants were assayed for infection of P4-CCR5 indicator cells. Infectivity was calculated after normalizing for the RT content of the SIV inocula. Values shown are normalized against the infectivity of the vehicle-treated virus stock. The absolute infectivity values of the control viruses are shown in Fig. 2D. The results are representative of two independent experiments.
